Oxidative phosphorylation is the final stage of cellular respiration, a process that cells use to produce energy. It occurs in the mitochondria, the powerhouse of the cell, where it uses the energy stored in the form of a hydrogen ion gradient across the mitochondrial membrane to generate ATP, the cell's main source of energy.
The process begins with the electron transport chain, a series of protein complexes located in the inner mitochondrial membrane. Electrons, which are derived from the breakdown of glucose in earlier stages of cellular respiration, are passed along these complexes. As the electrons move, they cause hydrogen ions (protons) to be pumped from the mitochondrial matrix into the intermembrane space, creating a gradient.
This gradient, also known as a proton motive force, is the key to ATP synthesis. The protons can only pass back into the mitochondrial matrix through a protein complex called ATP synthase. As they flow through ATP synthase, their potential energy is used to add a phosphate group to ADP, creating ATP.
The final electron acceptor in the electron transport chain is oxygen. Oxygen accepts the electrons and picks up protons to form water, a process known as reduction. This is why the process is called oxidative phosphorylation: it involves the transfer of electrons to oxygen (oxidation) and the addition of a phosphate group to ADP (phosphorylation).
