The pH of the environment in which an enzyme operates can significantly influence its activity. This is because pH affects the ionisation of the amino acids that make up the enzyme. A change in pH can alter the charge of these amino acids, which in turn can change the shape of the enzyme, including its active site. If the active site's shape is altered, the enzyme may no longer be able to bind to its substrate, or it may do so less efficiently, reducing its catalytic activity.
Moreover, each enzyme has an optimal pH at which it functions most efficiently. This optimal pH is often the pH of the enzyme's natural environment. For example, pepsin, an enzyme that breaks down proteins in the stomach, has an optimal pH of around 2, which is very acidic. In contrast, trypsin, an enzyme that also breaks down proteins but in the small intestine, has an optimal pH of around 8, which is slightly alkaline. If the pH deviates too far from this optimum, the enzyme's activity can decrease dramatically.
